1. Is Mandavi Express best to see Dudhsagar? Or I want to hire a car from Goa to see Dudhsagar?

2. How much time Mandavi express stay in front of Dudhsagar?

3. What will be the car cost from Anjuna Beach to Dudhsagar?

4. What time will be better to go from Mumbai to Goa via train? Day Or Night?

The Mandovi Express doesn't go anywhere near Dudhsagar. You need to look at trains between Madgaon and Londa Junction.

The Mumbai to Goa trip has some great scenery. I would try to do it during daylight hours

Yes, Mandovi exp doesn't go through Dudhsagar. Just board any train going towards Dudhsagar and get off at Dudhsagar station. Enjoy the waterfall and again board any train going to Madgaon or Londa where you want. All passing trains stop at Dudhsagar station. You may board any goods train from Kulem station to reach Dudhsagar and again catch any goods train to come back to Goa. Here is a huge goods train traffic.

To travel from Mumbai to madgaon , the best option is an early morning Shatabdi if you can wake up early in the morning. Or there are couple of overnite trains.

There is no Shatabdi from Mumbai to Madgaon. It is actually Janshatabdi (Train no. 12051).
Madgaon to Londa normally trains take 3 hours and Madgaon to Dudhsagar 2 hours.

The Goa Express which goes from via Londa will reach Dudhsagar and will stop for 1-2 minutes there. Only if it is delayed by a couple of hours then you will be able to see the waterfall. There are many agencies which arrange bike rides to the Dudhsagar. You have to set apart an entire day for this expedition.

There's no train which will actually stop at the waterfalls. You'll have to get down at Doodhsagar station and walk to the falls and back again to the station to catch a train. Doodhsagar is not a proper station. Trains stop here just for a routine brake checking if I'm not mistaken. An alternative would be to get down at Kulem and hire a 'pilot' (motorcycle) to take you to the falls. The approach road for cars and such is blocked during the monsoon by random streams, so only these pilots have an accessible route.

There are quite a few blogs on Doodhsagar. Read them up. Also check the train timings.
