You have to train or bus to Jodhpur, then train or bus to Ajmer for further transfer by bus to Pushkar. There is no direct train.
There is an overnight bus - Jaisalmer - Pushkar, but unless you are young, hale and hearty I wouldn't recommend it.
Every cloud has a silver lining!

There's a lot of travelling from Mumbai to Jaisalmer. I suggest you do this in stages, say, Mumbai to Surat - Surat to Ahmedabad (all by train) and from there take the overnight sleeper bus. Or else from Ahmedabad, train to Jodhpur and bus or overnight train,to Jaisalmer. Check out how long Mumbai to Ahmedabad train takes - I think a very long journey.

A good idea, haydn. There are a couple of trains going Mumbai to Ajmer, and many more Ahmedabad to Ajmer.

There are private and government buses leaving daily throughout the day from the bus stand in Pushkar for Jodhpur. From Jodhpur you can either take another bus to Jaisalmer or catch the evening train departing around 11.15 p.m. from Jodhpur railway station. Buses are going every hour for Jaisalmer from Bombay Motors area where the private bus stand is.

That's a good one VSP for anyone wanting to go all the way to Jaipur. And also the choice to get off at Ajmer for Pushkar. (Take the local bus - frequent departures and not expensive).
