Both provide bus services to Manali. HRTC buses leave from ISBT Kashmere Gate. I don't know where HPTDC buses leave from. Easiest thing would be to call them: http://hptdc.nic.in/bus.htm
This discussion happens every year on this thread. Guys, please read the posts from the previous year.

Hi Pooja DHPTDC Volvo buses have diiferent different pikup points like Kashmiri Gate, ISBT Bus stand, Cannought place. SO you have to fix it your own which one is comforatble for you. I used to take from ISBT bus Stand only.
Please look after that if you would travelling via HPTDC volvo then book ur ticket in advance because May would be the peak season for Manali. Get down at Bhunter and catch a bus for Manikaran which will drop you at Kasol Base camp.
