Can't tell you anything about the bus connection, but the train connection is very good. There is a daily night train from Madurai to Trivandrum, and travelling by train is much more relaxed than by road, be it in a luxury bus or a normal one...

Not much help maybe but I did the journey in January, by ordinary bus from the Central Bus Stand (didn't notice any luxury buses), and it was ok. We had to change in Nagercoil though which was slightly confusing and very hot. Took about 7 hours.

It's 264 km. to Trivandrum from Madurai.

www.madurai.com

has bus, train and air info but I saw nothing on a bus to Trivandrum.

Scroll down to the bottom of the front page of Madurai.com and you'll find other website's of towns in the area. They should have bus info also.
