you can plan your trip, train-wise, here http://www.irctc.co.in/
it will tell you the KMs between cities.
This March I took the train from Chennai to Madurai, then bus to Kodaikanal, then hired a car from Kodai to Rameswaram, then took train back to Chennai from Rameswaram. I spent about 2 days in each city, so spent a little over a week traveling, not 2 or 3 days total. The train from Chennai to Madurai is 9 hours; the train from Rameswaram to Chennai is 17 hours. The car ride from Kodai to Rameswaram was about 7 hours.
I recommend Park Hotel in Madurai first, Hotel Supreme second. Hotel Tamil Nadu in Rameswaram.
