Delhi - Tirupati - Rameshwaram - Madurai - Kanyakumari - Delhi
Not all could be covered by trains.
Few problems
Only one train from Tirupati to Rameshwaram (6734) on Thurdays
Only one train from Rameshwarm to Madurai (6733) on Fridays
Only two trains from Kanyakumari to Delhi (2641) on Wednesdays and (6317) on Fridays.
A few places would have to be covered by bus / taxi

Or after Tirupati, try making it Chennai, halt and plan. You can plan better with more travel options.

Madurai: College house, just opposite the railway station, walkable. if you don't like it or it's full, Town Hall road has more Hotels in the same place
Rameshwaram, Main temple street has lots, choose any one with A/C if you plan to stay for more than a day. Flies, there are lots of them. Auto fare from rail station to hotel will cost you about 40

Would second that.
Chennai is around 150 kms from Tirupati and there are 2 trains (6101) leaving at 19:55 and (6703) leaving at 17:00 running daily to Rameswaram

Delhi Tirupati 2626
TPTY Chennai 6054
MS Kanyakumari 2633
Evening train 2634 to Madurai. Or else other options from Nagercoil Junction
751SR to Rameswaram
6102 Chennai Express
Tamil Nadu Express to New Delhi

Shorter journeys of 3 - 4 hours can be covered by bus as well.

You should try and get to Tanjore, to see the awesome Brihadeshwara temple, a World Heritage site.
