1 night excursion from Manali
I am planning a trip to Manali and have booked hotel for 3 nights there. I have fourth night and hence about 1.5 days available. I am planning to go to Keylong for one night. Is it advisable to go to Keylong for one night. Lahaul and Spiti district website states travel time is 6 hour from Manali. Is it advisable to go by bus/cab? Though after reading about Keylong i would like to visit Keylong, but in case it is not advisable to visit Keylong for a night, what other place can I visit.

Keylong is not advised for one night while travelling from Manali. First, it is too beautiful to be devoted maybe just one evening & secondly, the trip would be too exhaustive. The drive may technically be 6 hours but one does devote more time at Rohtang.

i think you can squeeze in kasol in 1.5 days. takes about 5 hours to go to kasol from manali (via kullu and bhuntar). you could walk around kasol and have a picnic there somewhere and then come back to manali.
