Buses should be available from Manali bus stand. There are Himachal and J&K state transport buses for Leh. I highly doubt whether you can book them in advance unless you go there physically. In three day on that road I spotted only ordinary buses plying so don't expect luxury buses. The fare should be very reasonable though I don't have exact idea. I would expect around Rs. 200-300. This will take two days with one night stopover at Sarchu/Pang but journey will be treacherous.

There are jeeps which take you flat in 18 hours starting at 2AM but no fun and bone-jarring.

If you are a group then best hire an exclusive jeep. This way you will be flexible. You will get plenty of operators around Manali bus stand.

You can book your tickets to Leh up here: http://www.himachalhotels.in/

That would be for Himachal Road Transport Corp buses... and youd have a choice of a deluxe / Luxury bus

HPTDC buses tickets have to be bought physically in Manali or at one of their offices in one of the seven metros...

A ticket in an HRTC bus would cost roughly Rs. 425 per person, hiring a taxi exclusively would cost roughly 8-10k from Manali.
