Best is to hire a car and head north.
You'll get to mathura first and then to Vrindawan. They are enough to take your full day if still have time you can add visit to Goverdhan parvat (the hill which Krishna lifted on his finger to save from rains). Lot of people do Goverdhan parikrama on foot but in modern days you can do it by car

You can expect it to cost 1-1.5 k.
Btw, there is frequent and fast bus service between Agra and Mathura.
And entire local tour of Mathura / Brindavan (each separately) can be leisurely done on cycle rickshaws that may charge you 100 bucks for a couple of hours!
Tempo travelers are aplenty to reach Brindavan from Mathura.

If I stop over in Mathura for 1 HOUR on my way from Agra to Delhi by car, what are the main sights I can cover?

You should be able to visit the janma bhoomi and the couple of other important temples in the city.
Note that some are closed for 3-4 hours in the afternoon. Better you visit before 12 AM or after 4:30.
